AbstrakPenelitian ini dilakukan untuk mengetahui keberadaan bakteri pathogen menggunakan teknik isolasi dan deteksi Aeromonas hydrophila yang menginfeksi lele dumbo (Clarias gariepinus) di Kabupaten Banyumas dengan metode Polymerase Chain Reaction (PCR) menggunakan primer Aerofd dan AeroRs. Sampel lele dumbo (Clarias gariepinus) sakit diambil dari tiga daerah berbeda di Banyumas yaitu Pamijen, Rempoah dan Langgongsari. Bakteri diisolasi dan diekstraksi DNAnya untuk digunakan dalam PCR. Hasil penelitian menunjukkan gejala klinis lele dumbo (Clarias gariepinus) yang terserang bakteri pathogen adalah bercak merah pada sirip (punggung, ekor dan dada), sirip gripis, kulit terkelupas dan perut kembung. Delapan isolat bakteri pathogen berhasil didapatkan. Teknik PCR berhasil digunakan untuk mendeteksi Aeromonas hydrophila yang ditunjukkan fragmen DNA dengan ukuran 209 pasang basa pada gel agarose dan konfirmasi sekuen DNA menggunakan analisis BLAST. Hasil analisis BLAST menunjukkan isolat Aeromonas hydrophila dari Kabupaten Banyumas memiliki gen Aerolysin yang sekuennya mirip dengan sekuen gen Aerolysin Aeromonas hydrophila lain di Gen Bank. Empat dari delapan isolat terdeteksi sebagai Aeromonas hydrophila berdasarkan hasil PCR.
Abstrak (Inggris)The aims of this research were to determine occurrence of bacterial pathogen using isolation technique and to detect Aeromonas hydrophila which infected African catfish (Clarias gariepinus) in Banyumas Regency by using Polymerase Chain Reaction (PCR). Fish showed bacterial infection signs were collected from three different areas in Banyumas namely Pamijen, Rempoah and Langgongsari. Bacterial pathogen was isolated and its DNA sample was extracted to be used in PCR technique. Clinical symptoms of bacterial disease included red spots on fins (dorsal, caudal and pectoral), damaged fin, flaky skin and abdominal dropsy. Eight isolates of bacteria were collected. PCR techniques was successfully used to detect Aeromonas hydrophila which is indicated by specific DNA fragment of 209 base pairs on agarose gel and DNA sequence confirmation by using BLAST analysis. BLAST analysis results showed Aerolysin gene of Aeromonas hydrophila which was isolated from Banyumas is highly similiar to Aerolysin gene of Aeromonas hydrophila available in Gen Bank. Four of eight isolates were detected as Aeromonas hydrophila based on the PCR result.
