Judul ArtikelAPLIKASI METABOLIT SEKUNDER DUA ISOLAT Trichoderma harzianum TERHADAP PENYAKIT BERCAK DAUN (Pestalotia sp) PADA BIBIT TANAMAN MANGGIS
AbstrakPenelitian ini bertujuan untuk: (1) mengetahui daya hambat metabolit sekunder Trichoderma harzianum isolat jahe dan bawang merah maupun gabungannya terhadap pertumbuhan jamur patogen Pestalotia sp. in vitro; (2) mengetahui pengaruh aplikasi metabolit sekunder T. harzianum isolat jahe dan bawang merah, maupun gabungannya, terhadap penyakit bercak daun dan kandungan senyawa fenol tanaman, serta pertumbuhan bibit tanaman manggis. Penelitian ini dilaksanakan pada bulan Oktober 2016 sampai dengan Februari 2017 di Rumah Kaca dan Laboratorium Penyakit Tanaman Fakultas Pertanian Universitas Jenderal Soedirman. Penelitian in vitro menggunakan Rancangan Acak Lengkap (RAL) yaitu dengan 4 perlakuan yaitu kontrol, metabolit sekunder T. harzianum isolat jahe, bawang merah dan gabungan. Penelitian in planta menggunakan Rancangan Acak Kelompok (RAK) yaitu dengan 5 perlakuan dan 6 ulangan. Perlakuan terdiri atas kontrol, fungisida (berbahan aktif benomil), metabolit sekunder T. harzianum isolat jahe, isolat bawang merah, serta gabungan isolat jahe + bawang merah. Variabel yang diamati yaitu daya hambat secara in vitro, intensitas penyakit, luas bercak, tinggi tanaman, jumlah daun, luas daun baru, dan analisis jaringan. Hasil penelitian menunjukkan bahwa perlakuan metabolit sekunder T. harzianum isolat jahe dan bawang merah maupun gabungannya mampu menghambat pertumbuhan Pestalotia sp. sebesar 55,83%; 55,53%; dan 55,37% in vitro. Perlakuan metabolit sekunder T. harzianum gabungan isolat jahe + bawang merah mampu menekan intensitas penyakit bercak daun sebesar 80%, luas bercak 81,94% dan meningkatkan kandungan senyawa fenol (saponin, tanin dan glikosida) di dalam jaringan tanaman manggis, serta mampu meningkatkan pertumbuhan bibit tanaman manggis 28,6%, jumlah daun 86,82%, dan luas daun baru 21,35%.
Abstrak (Inggris) This study aimed to: (1) to acknowledge the inhibition of secondary metabolites of T. harzianum isolates ginger and shallot or a combination there of on the growth of pathogenic fungus Pestalotia sp. In vitro;(2) the effect of the application of secondary metabolites T. harzianum isolates ginger and shallot, or a combination there of, against leaf spot disease and plant phenolic compounds, and effect of secondary metabolites of two isolates of T. harzianum on the growth of mangosteen seedling. This research was conducted in October 2016 until February 2017 at the Greenhouse and Protection Plant Laboratory of the Faculty of Agriculture, Jenderal Sudirman University. In vitro uses a completely randomized design (CRD) that is by 4 ie control, secondary metabolites of T. harzianum and combined isolates ginger + shallot. Research in planta using randomized block design (RBD) in planta,namely with 5 treatments and 6 replications. The treatments consisted of control, fungicide (active ingredient benomil), secondary metabolites of T. harzianum isolates ginger, shallot and their combined ginger + shallot. Variables observed were inhibition in vitro, the intensity of the disease, spot area, plant height, leaf number, new leaf area, and tissue analysis. The results showed that treatment of secondary metabolites of T. harzianum isolates ginger and shallot or a combination of ginger + shallot isolates were able to inhibit the growth of Pestalotia sp. amounting to 55.83%; 55.53%; and 55.37% invitro.Treatment of secondary metabolites of T. harzianum combined isolate ginger + shallot able to suppress the intensity of leaf spot disease by 80%, broad patches of 81.94% and increase the content of phenolic compounds (saponins, tannins and glycoside) in the mangosteen plant tissue, and enhanced growth mangosteen seedlings was 28.6%, 86.82% the number of leaves and leaf area only reached 21.35%.
