| Abstrak | Periodontitis merupakan penyakit periodontal yang memiliki prevalensi cukup tinggi di masyarakat. Manifestasi klinisnya berupa kehilangan perlekatan dan tulang alveolar. Host modulating therapy dikembangkan sebagai metode perawatan pada periodontitis, salah satunya gel kitosan-klodronat. Penelitian ini bertujuan untuk mengetahui pengaruh aplikasi gel kitosan-klodronat secara intrapoket terhadap kadar osteocalcin pada tikus Sprague dawley periodontitis yang diinduksi bakteri A. antinomicetemcomitans. Jenis penelitian adalah eksperimental laboratoris dengan posttest only control group design. Tikus Sprague dawley sebanyak 35 ekor dibagi menjadi 5 kelompok: K1 (periodontitis, gel kitosan-klodronat 0,1%), K2 (periodontitis, gel kitosan-klodronat 0,2%), K3 (periodontitis, gel kitosan-klodronat 0,4%), K4 (kontrol negatif, gel carbopol 940), dan K5 (kontrol sehat). Kadar osteocalcin jaringan gingiva tikus diperiksa dengan uji ELISA. Hasil dianalisis menggunakan one way ANOVA dan dilanjutkan post hoc LSD. Rerata kadar osteocalcin pada kelompok K1 (7,980 ng/ml), K2 (8,845 ng/ml), K3 (10,024 ng/ml, K4 (6,686 ng/ml), dan K5 (7,475 ng/ml). Uji one way ANOVA menunjukkan terdapat perbedaan bermakna antar kelompok (p<0,05). Hasil uji post hoc LSD terdapat perbedaan antara K1, K2, K3 dengan K4, K2 dan K3 dengan K5, sedangkan K1 dengan K5 tidak terdapat perbedaan bermakna. Simpulan dari penelitian ini adalah kadar osteocalcin pada tikus Sprague dawley periodontitis akibat induksi bakteri Aggregatibacter actinomycetemcomitans pada aplikasi gel kitosan-klodronat 0,1%, 0,2%, dan 0,4% secara intrapoket lebih tinggi dibandingkan dengan gel carbopol 940. Kadar osteocalcin pada tikus Sprague dawley periodontitis pada aplikasi gel kitosan-klodronat 0,2% dan 0,4% lebih tinggi dibandingkan dengan kontrol sehat, sedangkan kadar osteocalcin pada aplikasi gel kitosan klodronat 0,1% sama dengan kontrol sehat. |
| Abstrak (Inggris) | Periodontitis is a periodontal disease that has a high prevalence in the community. Clinical manifestations include loss of attachment and alveolar bone. Host modulating therapy developed as a method of treatment in periodontitis, one of them are chitosan-clodronate gel. This study aims to determine the effect of chitosan-clodronate gel for intrapocket application on osteocalcin levels in Sprague dawley rats periodontitis due to bacterial induction Aggregatibacter antinomicetemcomitans. The type of research was experimental laboratory with posttest only control group design. Sprague dawley rats as many as 35 were divided into 5 groups: K1 (periodontitis, chitosan-clodronate gel 0,1%), K2 (periodontitis, chitosan-clodronate gel 0,2%), K3 (periodontitis, chitosan-clodronate gel 0,4 %), K4 (negative control, carbopol 940 gel), and K5 (healthy control). The levels of osteocalcin of rat gingival tissue were examined by ELISA assays. The results were analyzed using one way ANOVA and continued post hoc LSD. The mean levels of osteocalcin in the group K1 (7,980 ng / ml), K2 (8,845 ng / ml), K3 (10,024 ng / ml, K4 (6,686 ng / ml), and K5 (7,475 ng / ml). One way ANOVA showed there is a significant difference between groups (p<0,05). Post hoc LSD test there is a difference between K1, K2, K3 to K4, K2 and K3 with K5, while K1 to K5 are not significantly different. The conclusions of this study are the level of osteocalcin in Sprague dawley rats periodontitis due to bacterial induction Aggregatibacter antinomicetemcomitans on the intrapocket application of chitosan-clodronate gel 0,1%, 0,2%, and 0,4% higher than carbopol 940 gel. The levels of osteocalcin in Sprague dawley rats periodontitis on the application of chitosan-clodronate gel 0,2% and 0,4% higher than healthy controls, whereas the levels of osteocalcin in chitosan-clodronate gel 0,1% is equal to healthy controls. |
