Judul ArtikelPENGARUH METODE THAWING DAN LAMA POST THAWING TERHADAP INTEGRITAS MEMBRAN DAN ABNORMALITAS SPERMATOZOA SAPI SIMMENTAL
AbstrakPenelitian ini bertujuan untuk mengetahui interaksi metode thawing dan lama post thawing terhadap integritas membran serta abnormalitas spermatozoa sapi Simmental. Materi penelitian yang digunakan adalah 24 straw sapi Simmental berasal dari Dinas Peternakan dan Perikanan Kabupaten Banyumas, larutan eosin negrosin 50 ml, 24 ml larutan Hipoosmotik Swelling Test (HOS Test) (1,351 g fruktosa, 0,735 g Na-sitrat, 100 ml Aquabidest). Semen beku di thawing menggunakan air suhu 37oC selama 30 detik dan air PDAM selama 30 detik dengan lama post thawing 20, 40 dan 60 menit. Metode penelitian menggunakan Rancangan Acak Lengkap (RAL) pola factorial dengan 6 perlakuan dan 4 ulangan. Peubah yang diukur adalah integritas membran dan abnormalitas spermatozoa sapi Simmental. Hasil penelitian menunjukan tidak ada interaksi antara metode thawing dan lama post thawing (P>0,05) terhadap integritas membran dan abnormalitas sapi Simmental. Akan tetapi, lama post thawing berpengaruh nyata (P<0.05) menurunkan persentase integritas membran dan menaikan persentase abnormalitas spermatozoa sapi Simmental. Rataan persentase integritas membran dan abnormalitas pada metode thawing air suhu 37oC adalah 44,56 ± 1,8% dan 9,73 ± 1,4% sedangkan pada metode thawing air PDAM adalah 27,12 ± 2,0% dan 11,02 ± 2,1%. Rataan persentase integritas membran pada lama post thawing 20, 40, 60 menit adalah 40,16 ± 2.8%; 35.00 ± 1.0%; 32.36 ± 1.4%. Sedangkan rataan persentase abnormalitas pada lama post thawing 20, 40, 60 menit adalah 8,80 ± 0,9%; 10,35 ± 1,7%; 11,96 ± 2,3%. Berdasarkan hasil penelitian metode thawing menggunakan air suhu 37oC selama 30 detik dengan lama post thawing 20 menit memperoleh kualitas semen yang baik dan layak digunakan untuk keperluan inseminasi buatan.
Abstrak (Inggris)The purpose of this research was to find out the interaction between thawing method and post thawing time on membrane integrity and Simmental spermatozoa abnormality. The research material used was 24 straw Simmental cattle from Livestock and Fishery Department of Banyumas Regency, 50 ml eosin nigrosin solution, 24 ml of Hipoosmotic Swelling Test (HOS Test) (1.351 g fructose, 0.735 g Na-citrate, 100 ml Aquabidest). The frozen semen was thawed using 37oC temperature water for 30 seconds and tap water for 30 seconds with post thawing length of 20, 40 and 60 minutes. Research method using Completely Randomized Design (CRD) factorial pattern with 6 treatments and 4 replicates. The measured variables were Simmental’s spermatozoa membrane integrity and spermatozoa morphological abnormality. The results showed that there was no interaction between thawing method and post thawing time (P>0.05) on Simmental’s spermatozoa membrane integrity and spermatozoa morphological abnormality. However, post thawing duration significantly (P<0.05) decreased the percentage of membrane integrity and increased the percentage of Simmental cattle spermatozoa abnormalities. The percentage of membrane integrity and abnormality in thawing method of temperature 37oC was 44.56 ± 1.8% and 9.73 ± 1.4% while the tap water thawing method was 27.12 ± 2.0% and 11.02 ± 2.1%. The mean percentage of membrane integrity at post thawing length of 20, 40, 60 min was 40.16 ± 2.8%; 35.00 ± 1.0%; 32.36 ± 1.4%. While the mean percentage of abnormality in different post thawing time was 8.80 ± 0.9%; 10.35 ± 1.7%; 11.96 ± 2.3%. Based on thawing method, it uses 37oC temperature for 30 second with 20 minutes post thawing time to get good semen quality and suitable for artificial insemination.
