AbstrakPenelitian ini dilakukan untuk mengetahui aktivitas amilase organ pencernaan ikan uceng pada pH dan temperatur inkubasi yang berbeda serta mengetahui pH dan temperatur inkubasi optimum aktivitas amilase pada organ pencernaan ikan uceng. Penelitian ini terdiri dari dua tahap yang dilakukan secara eksperimental dengan menggunakan metode penelitian Rancangan Acak Lengkap (RAL) dan Rancangan Acak Lengkap (RAL) pola faktorial 2x3. Penelitian tahap pertama dilakukan untuk mengetahui aktivitas amilase organ pencernaan ikan uceng pada enam pH buffer yang berbeda (1,7, 3,4, 5, 7, 8, dan 10) dan setiap perlakuan diulang empat kali. Penelitian tahap kedua yaitu untuk mengetahui aktivitas amilase organ pencernaan ikan uceng pada tiga temperatur inkubasi enzim yang berbeda (30 ºC, 40 ºC, dan 50 ºC) dan setiap perlakuan diulang empat kali. Aktivitas amilase diukur dengan metode spektrofotometri. Hasil penelitian menunjukkan bahwa perbedaan pH dan temperatur inkubasi memberikan pengaruh nyata (p<0,05) terhadap aktivitas amilase organ pencernaan ikan uceng. Aktivitas amilase optimal diperoleh pada inkubasi pH 7 hingga pH 8 dan temperatur 30 ºC hingga 40 ºC.
Abstrak (Inggris)Uceng fish (Nemacheilus fasciatus) is one of wild fish that has not been cultivated and very potential to be developed. Currently the existence of uceng fish in nature is very rarely because of the utilization by humans to be consumed and traded as ornamental fish. This condition triggers the need for conservation of the fish to keep its existence. Conservation need to be supported by the knowledge of fish biology. Initial studies of fish have been done, but still limited to ecological and taxonomic aspects. A physiological study of digestion capacity reflected as a digestive enzyme activity is essential to determine the efficiency of nutrient utilization in fish in order to support fish conservation. The purpose of this research is to know the effect of pH and incubation temperature on amylase activity on digestive organ of uceng fish and to know pH and optimum incubation temperature of amylase activity in digestive organ of uceng fish. This research consists of two stages which will be done experimentally by using Completely Randomized Design (CRD) and Completely Randomized Design (CRD) with 2 x 3 factorial design. The first stage of the research was conducted to find out the activity of amylase of digestive organ of uceng fish at six different buffer pH (1,7, 3,4, 5, 7, 8, and 10) and each treatment was repeated four times. The second phase of the research was to know the activity of amylase of digestive organ of uceng fish at three different enzyme incubation temperature (30 ºC, 40 ºC, and 50 ºC) and each treatment was repeated four times. Amylase activity was measured by spectrophotometric method. The results showed that the difference of pH and incubation temperature gave a real effect (p <0,05) to amylase activity of digestive organ of uceng fish. Optimal amylase activity was obtained at incubation pH 7 to pH 8 and temperature 30 ºC to 40 ºC.
