Judul ArtikelKARAKTERISASI MORFOLOGI BAKTERI TANAH ULTISOL DAN PERTUMBUHANNYA PADA BEBERAPA KONSENTRASI PESTISIDA BERBAHAN AKTIF BUPROFEZIN
AbstrakPenelitian ini bertujuan untuk : 1) mengisolasi dan mengkarakterisasi morfologi bakteri rhizosfer tanah ultisol, 2) menguji kemampuan pertumbuhan bakteri tanah ultisol pada 4 perlakuan yakni kontrol, konsentrasi 5 ppm, 10 ppm dan 15 ppm pestisida berbahan aktif buprofezin. Penelitian ini dilaksanakan pada bulan April 2017 sampai November 2017 di Laboratorium Riset, Laboratorium Agroekologi, Laboratorium Agronomi, Laboratorium Perlindungan Tanaman dan Laboratorium Pemuliaan Tanaman, Fakultas Pertanian, Universitas Jenderal Soedirman. Penelitian ini diawali dengan pengambilan sampel tanah ultisol dari 4 lokasi yang berbeda Desa Srowot, Desa Pagaralang, Desa Karangrao dan Desa Tanggeran Kabupaten Banyumas. Sampel tanah diisolasi dan diidentifikasi bakteri yang tumbuh, bakteri terbaik diuji kemampuan pertumbuhannya pada 3 konsentrasi pestisida buprofezin 5 ppm, 10 ppm dan 15 ppm. Variabel yang diamati seperti populasi bakteri (TPC), morfologi, pertumbuhan selama 24 jam, uji pewarnaan gram, uji pertumbuhan dengan 4 perlakuan dan pH medium kultur bakteri. Hasil penelitian yang diperoleh dari seleksi isolasi didapatkan 4 isolat bakteri rhizosfer tanah ultisol yaitu isolat SR2 bakteri gram positif (koloni bundar, berwarna putih, batas tepi licin, ukuran kecil) , TG4 bakteri gram negatif (koloni bundar, berwarna putih, batas tepi licin, ukuran kecil), KR1bakteri gram negatif (koloni bundar,transparan, batas tepi tipis namun timbul, ukuran kecil) dan PA11 bakteri gram negatif (koloni bundar, berwarna putih, batas tepi tak beraturan, ukuran koloni kecil). Isolat bakteri SR2 dapat tumbuh baik pada perlakuan kontrol dan konsentrasi 5 ppm pestisida berbahan aktif buprofezin..
Abstrak (Inggris)This research aims to: 1) isolate and characterize bacteria morphology on ultisol soil, 2) tested the capability of ultisol soil bacteria growth on four treatments there is control, the concentration 5ppm, 10 ppm, and 15 ppm on pesticide buprofezin. The research was conducted in April until November 2017 at Research, Agroecology, Agronomy, Plants Protection, and Breeding Laboratory, Faculty of Agriculture, Jenderal Soedirman University. This research begins with ultisol soil sampling from four different locations are Srowot, Pagaralang, Karangrao, and Tanggeran Village, Banyumas Regency. Soil samples were isolated and identified by bacteria growth, the finest bacteria would be tested for growth ability on three Buprofezin concentrations are 5 ppm, 10 ppm, and 15 ppm. The variables observed with the bacterial population (TPC), morphology test, growth for 24 hours test, gram staining test, growth on 4 pesticide concentration test, and bacterial culture medium acidity (pH). The results showed that 4 selected soil were Srowot samples or isolate SR2 included with gram-positive bacteria (round colonies, white color, slippery border, and small size), Tanggeran sample or TG4 included with gram-negative (round colonies, white color, slippery border, and small size), Karangrao samples or isolate KR1 included with gram-negative (circular colonies, transparent colors, thin but occurring margins, and small size) and Pageralang samples or isolate PA11 included with gram-negative (round colonies, white color, irregular, and small size). Isolates SR2 can growth well and adapted to control and 5 ppm of Buprofezin concentrations treatment.
