AbstrakKalium diklofenak adalah obat golongan antiinflamasi nonsteroid (AINS) yang sering digunakan sebagai terapi analgesik pada kondisi inflamasi maupun nyeri pada penyakit Osteoarthritis dan Rheumatoid Arthritis. Kalium diklofenak dibuat dalam bentuk tablet dispersibel yang penggunaannya didispersikan terlebih dahulu ke dalam larutan, agar onset tercapai lebih cepat dan mudah digunakan, terutama bagi pasien yang kesulitan menelan tablet secara utuh. Tujuan penelitian ini untuk mengetahui pengaruh kombinasi dan proporsi optimum campuran Explotab dan Avicel pH-101 dalam formulasi tablet dispersibel Kalium diklofenak dengan metode Simplex Lattice Design 2 faktorial. Formula tablet berdasarkan metode SLD dengan eksipien Explotab (A) dan Avicel pH-101 (B) yaitu: FI (100%A), FII (75%A:25%B), FIII (50%A:50%B), FIV (25%A:75%B), dan FV (100%B). Tablet dibuat dengan metode kempa langsung. Kemudian diuji sifat fisiknya yang meliputi kekerasan, kerapuhan, keseragaman ukuran, keseragaman bobot, waktu hancur, waktu terdispersi, waktu terbasahi, dan uji penetapan kadar. Berdasar hasil penelitian didapatkan pengaruh kombinasi Explotab dan Avicel pH-101 pada sifat fisik tablet, yaitu menurunkan nilai keseragaman bobot, meningkatkan kekerasan tablet, menurunkan kerapuhan tablet, dan memperlama waktu hancur serta waktu terdispersi tablet dispersibel Kalium diklofenak. Formula optimum tablet dispersibel Kalium diklofenak didapat pada campuran 40% Explotab dan 60% Avicel pH-101. Pemilihan formula optimum didasarkan pada perhitungan nilai respon total yang tertinggi menggunakan metode Simplex Lattice Design 2 Faktorial. Abstrak (Inggris)A Potassium diclofenac is a non-steroidal anti-inflammatory (NSAID) drug that are frequently used as an analgesic in inflammatory and pain of Osteoarthritis as well as Rheumatoid Arthritis diseases. In the term of this research, the potassium diclofenac is made in a dispersible tablet, which by the use of the tablet is previously dispersed through a liquid, in order to attain a faster onset and easier to use, particularly to the patients who cannot swallow a whole tablet. The research aims to determine the combination impacts and the optimum proportion of Explotab also Avicel pH-101 in the formulation of the Potassium diclofenac dispersible tablet by doing a Simplex Lattice Design 2 factorial. The tablet formulation based on the SLD method with Explotab (A) and Avicel pH-101 (B) excipients as follows: FI (100%A), FII (75%A:25%B), FIII (50%A:50%B), FIV (25%A:75%B) and FV (100%B). The tablet is made by a direct compression method, and then evaluated for hardness, friability, size uniformity, weight variation, dispersion time, wetting time, disintegration time and determination of drug amount. According to the result of the research, the combination of Explotab and Avicel pH-101 have an impact on the physical properties of the tablet such as decreasing the weight uniformity and the fragility of tablet, increasing the hardness, lengthening the disintegration time and dispersed time of dispersible Potassium diclofenac tablet. The formula containing 40% Explotab and 60% Avicel pH-101 were consideribly selected as the optimum formula. Its determination is based on the calculation of the maximum total response value by doing the Simplex Lattice Design 2 factorial method.
