Judul ArtikelTOKSISITAS SUBKRONIK EKSTRAK ETANOL DAUN JARAK PAGAR (Jatropha curcas) TERHADAP FUNGSI GINJAL TIKUS WISTAR
AbstrakDaun jarak pagar (Jatropha curcas) terbukti memiliki aktivitas antiinflamasi. Uji toksisitas akut menunjukkan bahwa daun J. curcas aman digunakan. Penelitian ini bertujuan untuk mengetahui toksisitas subkronik daun J. curcas terhadap kondisi klinis, kadar kreatinin serum dan gambaran histopatologi ginjal tikus Wistar. Penelitian ini merupakan eksperimental murni dengan rancangan Post Test Only Controlled Group. Subjek uji 40 ekor tikus Wistar jantan dan betina dibagi acak menjadi 4 kelompok. Kelompok I adalah kontrol Na-CMC 1%. Kelompok II, III, dan IV diberikan ekstrak etanol daun J. curcas dengan dosis masing-masing 150, 300, dan 600 mg/kgBB secara peroral sekali sehari selama 30 hari. Hasil pengamatan gejala toksik klinis dianalisis deskriptif. Data kadar kreatinin serum dianalisis menggunakan uji one-way ANOVA. Sedangkan gambaran histopatologi ginjal setelah nekropsi hari ke-31 dianalisis deskriptif dan semikuantitatif dengan sistem skoring. Hasil penelitian menunjukkan bahwa ekstrak etanol daun J. curcas pada dosis 150, 300 dan 600 mg/kgBB tidak menyebabkan kematian maupun gangguan sistem SSP-somatomotor hewan uji, namun menimbulkan efek toksik pada saluran cerna berupa tinja hitam dan berlendir. Ekstrak dosis 150 mg/kgBB dinilai aman karena menimbulkan gejala toksik tinja hitam paling singkat dibandingkan kelompok kontrol. Ekstrak dosis 150 mg/kgBB juga terbukti aman terhadap organ ginjal karena tidak meningkatkan kadar kreatinin serum hewan uji diluar rentang nilai normal serta hanya menimbulkan perubahan struktur ginjal yang reversible berupa degenerasi parenkimatosa. Ekstrak dosis 300 dan 600 mg/kgBB tidak meningkatkan kadar kreatinin serum hewan uji hingga nilai yang menunjukkan gangguan fungsi ginjal, namun ditemukan kongesti dan nekrosis yang telah mengindikasikan terjadinya toksisitas ginjal secara struktural.
Abstrak (Inggris)Jatropha curcas leaves have been proven as anti-inflammatory agent. The Acute toxicity test showed that J. curcas leaves was safe to use. This study aimed to determine the subchronic toxicity effect of J. curcas leaves extract on clinical condition, serum creatinine level and kidneys histopathology appearances of Wistar rats. This research was an experimental study using the post test only controlled group design. Samples were 40 male and female Wistar rat, randomly divided into 4 groups. Group I was control with Na-CMC 1%, Group II, III, and IV were given ethanolic extract of J. curcas leaves with dose of 150, 300 and 600 mg/kgBW, respectively, while the extract was given orally once a day for 30 days. Clinical sign of toxicity were analyzed descriptively. Serum creatinine level was analyzed using one-way ANOVA test. While histopathology examination from necropsy on day 31 were analyzed descriptively followed by scoring system analysis. The result showed that J. curcas leaves ethanolic extract with dose of 150, 300 and 600 mg/kgBW didn’t caused a mortality and disorder in SSP-Somatomotor system, but it caused a gastrointestinal toxic symptoms such a dark and watery stool in animal test. The extract at 150 mg/kgBW was safe to use since it caused slightest dark-stool toxic symptoms compared to control group. Extract at 150 mg/kgBW also safe to the kidneys since it not made any alteration in serum creatinine level and just made a reversible lesion such a parenkimatosa degeneration on a kidneys histopatology. Extract at 300 and 600 mg/kgBW didn’t make any increasement in serum creatinine level which indicate the kidneys impairment, but the histopatological examination shows a congestion and necrosis that indicate already a kidneys toxicity.
