| Abstrak | Sodium diklofenak termasuk salah satu senyawa obat golongan non steroidal anti-inflammatory drugs (NSAIDs) yang dapat mengobati radang, rasa sakit, demam, encok, migrain dan lain-lain. Pada penelitian ini dilakukan validasi metode analisis untuk senyawa sodium diklofenak menggunakan kromatografi cair kinerja tinggi (KCKT). Sistem KCKT dioptimasi menggunakan kolom CYCLOBOND; komposisi fasa gerak acetonitril:air (60:40); laju alir 0,3 mL/menit; volume injeksi 5 µl dan dideteksi UV pada 282 nm. Kurva kalibrasi linear dalam rentang 5 – 40 ppm dengan nilai koefisien korelasi 0,9995. Batas deteksi dan batas kuantifikasi yang diperoleh masing-masing adalah 4,7 dan 15,7 ppm. Kombinasi SPE LC-18 dan metode KCKT digunakan dalam penentuan sodium diklofenak dalam sampel urine. Rata-rata % perolehan kembali (recovery) diperoleh sebesar 90,55% (RSD 1,17 % dan HORRAT 0,1). |
| Abstrak (Inggris) | Sodium diclofenac is a non-steroidal anti-inflammatory drugs (NSAIDs) that can treat inflammation, pain, fever, gout, migraine and others. In this study, validation of the method for the sodium diclofenac analysis was performed using high performance liquid chromatography (HPLC). The HPLC system is optimized using the CYCLOBOND column; composition of mobile phase acetonitrile: water (60:40); flow rate of 0.3 mL / min; injection volume of 5 μl and detected by UV at 282 nm. Linear calibration curves obtained in the range of 5 - 40 ppm with a coefficient correlation (r) 0.9995. Limit of detection (LOD) and limit of quantitation (LOQ) were obtained 4.7 and 15.7 ppm respectively. Combination of SPE using LC-18 and optimized HPLC method applied for determination of sodium diclofenac in a urine sample. Averange recovery is obtained of 90.55% (RSD 1.17% and HORRAT 0.1). |
