AbstrakPenyakit layu bakteri merupakan salah satu penyakit penting pada tanaman tomat yang disebabkan oleh Ralstonia solanacearum. Metabolit sekunder bakteri antagonis Pseudomonas fluorescens P60 berpotensi diuji dalam formula butiran untuk mengendalikan bakteri patogen R. soalanacearum penyebab penyakit Layu bakteri.Penelitian bertujuan mengetahui antagonisme dan daya simpan metabolit sekunder P. fluorescens P60 formula butiran, pengaruh aplikasi metabolit sekunder P. fluorescens P60 formula butiran terhadap penyakit layu bakteri dan pertumbuhan tanaman tomat di lapangan terbatas. Penelitian dilaksanakan di Laboratorium Perlindungan Tanaman dan Experimental farm Fakultas Pertanian Universitas Jenderal Soedirman dari bulan November 2020 sampai Agustus 2021. Penelitian terdiri dari dua tahap yaitu in vitro dan uji lapangan terbatas. Penelitian in vitro meliputi pengujian antagonisme dan daya simpan formula butiran P. fluorescens P60. Penelitian uji lapangan terbatas menggunakan Rancangan Acak Kelompok dengan enam perlakuan dan empat ulangan. Perlakuan yang dicoba yaitu R. solanacearum, R. solanacearum + 1, 5, 10, dan 15 g butiran, serta bakterisida (Agrimycin sulfat 20%). Variabel yang diamati meliputi zona hambat, masa inkubasi, intensitas penyakit, nilai Area Under Disease Progress Curve (AUDPC), tinggi tanaman, bobot segar tajuk, bobot segar akar, bobot kering tajuk dan bobot kering akar. Hasil penelitian in vitro menunjukkan formula butiran sampai dengan minggu ke-4 masih mempunyai aktivitas antagonisme yang baik dan memiliki penekanan penurunan zona hambat antara 21,67-23,34%. Hasil penelitian uji lapangan terbatas menunjukkan perlakuan metabolit sekunder P. fluorescens P60 formula butiran sebanyak 15 g efektif menurunkan intensitas penyakit sebesar 75%, menurunkan nilai AUDPC 74,72%, meningkatkan tinggi tanaman sebesar 44%, meningkatkan bobot kering akar sebesar 62,5%, meningkatkan bobot segar tajuk 65,65%. Perlakuan metabolit sekunder P. fluorescens P60 formula butiran mampu meningkatkan kandungan senyawa fenol secara kualitatif yaitu saponin, tanin, dan glikosida.
Abstrak (Inggris)Bacterial wilt is one of the tomato important diseases caused by Ralstonia solanacearum. Control the disease can be done by application of antagonistic bacteria secondary metabolites such as Pseudomonas fluorescens P60 in granular formulation. Aim of this study was to determine the antagonism and shelf life of P. fluorescens P60 secondary metabolite granular formula, the effect of P. fluorescens P60 granulated secondary metabolites application on bacterial wilt and on growth of tomato plants in the limited field. The research was conducted at the Plant Protection Laboratory and Experimental Farm, Faculty of Agriculture, Jenderal Sudirman University from November 2020 to August 2021. The research consisted of two stages, namely in vitro and limited field testing. In vitro test included testing the antagonism by storage time of the granular formula. Limited field test using a Randomized Block Design with six treatments and four replicates. The treatments were R. solanacearum without granules, R. solanacearum + 1, 5, 10, and 15 g granules, and bactericide (agrimycin sulphate 20%). Variables observed included inhibition zone, incubation period, disease intensity, Area Under Disease Progress Curve (AUDPC), plant height, crown fresh weight, root fresh weight, shoot dry weight, and root dry weight. Results of the research showed that the granular formula up to the 4th week had good antagonistic activity and able to suppress the bacterial growth between 21.67-23.34% compared to control. The treatment of granulated secondary metabolites of P. fluorescens P60 for 15 g effectively reduced the disease intensity and AUDPC as 75%, respectively, increase plant height, crown fresh weight, and root dry weight as 44, 65.65, and 62.5%, respectively, compared to control. The granulated secondary metabolites of P. fluorescens P60 was able to increase the content of phenolic compounds qualitatively, namely saponins, tannins, and glycosides.
