AbstrakRhizobacteria merupakan bakteri yang habitatnya pada tanah di sekitar perakaran yang dapat berasosiasi dengan akar tanaman. Asosiasi antara Rhizobacteria dan akar tanaman mampu meningkatkan pertumbuhan tanaman dan ketahanan tanaman melalui produksi fitohormon IAA (Indole Acetyc Acid) pada akar dan meningkatkan induksi ketahanan sistemik (ISR) pada tanaman. Penelitian ini bertujuan untuk mengetahui pengaruh inokulasi PGPR Indigenous lahan salin dalam mengendalikan penyakit hawar daun bakteri padi secara in vitro, in planta dan biokimia tanaman padi. Penelitian ini dilaksanakan pada Desember 2020 sampai Juli 2021 di Laboratorium Agronomi dan Holtikultura dan Screen House Experimental Farm Fakultas Pertanian Universitas Jenderal Soedirman. Penelitian ini menggunakan menggunakan Rancangan Acak Kelompok (RAK) faktor tunggal dengan 9 taraf perlakuan dan diulang 3 kali. Perlakuan pada penelitian ini meliputi kontrol tanpa inokulasi pgpr dan inokulasi Xoo dan inokulasi isolat JA1, JB1, JB2, JD1, JE1, R08, R011 yang inokulasi Xoo pada media AB Mix dengan tingkat salinitas air (DHL) 5,5 dSm^(-1). Hasil penelitian ini menunjukkan bahwa inokulasi isolat Rhizobacteria indigenous lahan salin mampu menghambat Xoo secara in vitro. Isolat Rhizobacteria indigenous JD1 memilik zona hambat sebesar 25,0 dengan daya hambat sangat kuat, Sedangkan JB2 dan JA2 termasuk dalam kategori yang kuat. Isolat PGPR indigenous lahan salin tidak mampu menunda masa inkubasi, kejadian penyakit dan intensitas penyakit, sedangkan isolat R08 dan JD1 mampu menurunkan nilai AUDPC dengan nilai masing-masing sebesar 24,34% dan 24,98% jika dibandingkan dengan perlakuan kontrol. Isolat PGPR indigenous lahan salin tidak mampu meningkatkan kandungan peroksidasi dan fenol total, akan tetapi mampu meningkatkan kandungan tanin dan saponin yang lebih baik dibandingkan perlakuan kontrol.
Abstrak (Inggris)Rhizobacteria are bacteria whose habitat is in the soil around the roots that can be associated with plant roots. The association between Rhizobacteria and plant roots can increase plant growth and plant resistance through the production of phytohormones IAA (Indole Acetyc Acid) in roots and increase the induction of systemic resistance (ISR) in plants. This study aims to determine the effect of PGPR Indigenous saline inoculation in controlling rice bacterial leaf blight in vitro, in planta and biochemically in rice plants. This research was conducted from December 2020 to July 2021 at the Laboratory of Agronomy and Horticulture and the Screen House Experimental Farm, Faculty of Agriculture, Jenderal Sudirman University. This study used a single factor Randomized Block Design (RAK) with 9 treatment levels and repeated 3 times. The treatments in this study included control without pgpr inoculation and Xoo inoculation and inoculation of isolates JA1, JB1, JB2, JD1, JE1, R08, R011 which Xoo inoculated on AB Mix media with a water salinity level (DHL) of 5.5 dSm^(-1 ). The results of this study showed that the inoculation of Rhizobacteria isolates indigenous to saline soil was able to inhibit Xoo in vitro. The indigenous Rhizobacteria isolate JD1 had an inhibition zone of 25.0 with a very strong inhibitory power, while JB2 and JA2 were included in the strong category. Indigenous PGPR isolates from saline land were not able to delay the incubation period, disease incidence and disease intensity, while R08 and JD1 isolates were able to reduce the AUDPC values by 24.34% and 24.98% respectively when compared to the control treatment. The indigenous PGPR isolate from saline soil was not able to increase the total peroxidation and phenol content, but was able to increase the tannin and saponin content which was better than the control treatment.
