Judul ArtikelUji Perbanyakan dan Lama Simpan Jamur Patogen Gulma dalam Limbah Cair Tempe dan Limbah Cair Tapioka, serta Aplikasinya pada Babadotan
AbstrakPenelitian bertujuan untuk mengetahui medium cair terbaik untuk perbanyakan jamur patogen gulma, lama simpan jamur pada medium, serta virulensi jamur patogen hasil perbanyakan. Penelitian dilaksanakan di Laboratorium Perlindungan Tanaman dan di Rumah Kaca, Fakultas Pertanian, Universitas Jenderal Soedirman, mulai bulan Oktober 2021 hingga Januari 2022. Penelitian terdiri atas dua tahap, tahap I uji perbanyakan jamur patogen gulma pada medium cair alternatif dengan rancangan acak lengkap faktorial, dengan faktor pertama jenis jamur patogen gulma (Curvularia sp., Fusarium sp., Chaetomium sp.) dan faktor kedua jenis medium cair (limbah cair tempe atau tapioka) dengan empat kali ulangan. Penelitian tahap II, uji in vivo pada gulma babadotan, dengan rancangan acak kelompok yang terdiri atas kontrol, limbar cair tempe + Chaetomium sp., Fusarium sp., atau Curvularia sp., dan limbah cair tapioka + Chaetomium sp., Fusarium sp., atau Curvularia sp., dengan empat kali ulangan. Variabel yang diamati kerapatan konidium jamur patogen, jumlah koloni, masa inkubasi dan gejala, intensitas penyakit, AUDPC, tinggi tanaman, jumlah daun, bobot tajuk segar, bobot tajuk kering, bobot akar segar, dan bobot akar kering. Kedua medium cair sesuai sebagai medium perbanyakan jamur patogen. Kerapatan konidium Curvularia sp. sebesar 28,42% lebih tinggi dibanding Fusarium sp. dan 99,9% lebih tinggi dibanding Chaetomium sp. Lama simpan terbaik untuk jamur patogen yaitu 6 minggu. Perlakuan medium limbah cair tempe + Curvularia sp. merupakan perlakuan terbaik, memiliki nilai masa inkubasi 3,33 hsi, nilai intensitas penyakit 88,78%, dan nilai AUDPC 713,25%-hari, tetapi bobot tajuk segar dan kering serta bobot akar segar dan kering tidak berbeda nyata.
Abstrak (Inggris)This research aims to determine the best liquid medium for propagation of weed pathogenic fungi, the duration of storage of the fungus on the medium, and the virulence of the multiplied pathogenic fungi. The research was carried out at the Plant Protection Laboratory and in the Greenhouse, Faculty of Agriculture, Jenderal Sudirman University, from October 2021 to January 2022. The study consisted of two stages, the first stage was testing the propagation of weed pathogenic fungi in alternative liquid medium with a factorial completely randomized design, with the first factor was the type of weed pathogenic fungus (Curvularia sp., Fusarium sp., Chaetomium sp.) and the second factor was the type of liquid medium (temperature or tapioca liquid waste) with four replications. Phase II research, in vivo on goatweed, with a randomized block design consisting of control, tempe + Chaetomium sp., Fusarium sp., or Curvularia sp. liquid waste, and tapioca + Chaetomium sp., Fusarium sp.or Curvularia sp., with four replications. Variables observed were conidium density of pathogenic fungi, number of colonies, incubation period and symptoms, disease intensity, AUDPC, plant height, number of leaves, fresh crown weight, dry crown weight, fresh root weight, and dry root weight. Both liquid media are suitable as a medium for propagation of pathogenic fungi. Conidium density Curvularia sp. by 28.42% higher than Fusarium sp. and 99.9% higher than Chaetomium sp. The best shelf life for pathogenic fungi is 6 weeks. Tempe treatment medium liquid waste + Curvularia sp. is the best treatment, has an incubation period of 3.33 hsi, a disease intensity value of 88.78%, and an AUDPC value of 713.25%-day, but the weight of fresh and dry shoots and the weight of fresh and dry roots were not significantly different.
